Yes—but a BSL-2 laboratory cannot become a BSL-3 facility by changing a sign or adding a filter. A workable conversion strategy designs the containment suite, mechanical systems, security, utilities, waste handling and operating procedures for both modes from the beginning. The laboratory can then run routine work at BSL-2 and activate BSL-3 controls when a protocol, agent and risk assessment justify them.
This approach can avoid paying the operating burden of full BSL-3 conditions for every experiment, but the financial case remains an architectural hypothesis rather than a demonstrated payback. A 2024 Building Design + Construction article by Mike Moreland and Ross Ferries of Flad proposes the model; it reports no project cost comparison, energy study, conversion duration or savings figure. The CDC/NIH Biosafety in Microbiological and Biomedical Laboratories (BMBL), sixth edition, supplies the governing context: design is driven by protocol-specific risk assessment and validation, not by a universal conversion recipe.

What a BSL conversion actually involves
In this context, “conversion” means planning a laboratory that can support two operating states. In ordinary use, the suite functions as BSL-2 space. When an approved protocol requires BSL-3 containment, the institution activates the additional controls, verifies the systems and restricts the area to the higher-containment workflow.
The distinction matters because BSL-3 is a complete operational system. Entry controls, room pressure relationships, directional airflow, decontamination, waste treatment, emergency procedures, training and maintenance must work together. A room that has enough square footage, a biological safety cabinet or a nominally suitable exhaust fan is not automatically BSL-3-capable.

The BMBL sixth edition is advisory guidance rather than a universal regulatory blueprint. Its foreword states: “The BMBL is not intended to be a regulatory document, although we recognize that some may use it in that way.” Local authorities, institutional biosafety committees, occupational-health programs, funders and other regulators can impose additional requirements.
Why institutions consider an adaptable suite
A dedicated BSL-3 facility is operated, maintained and secured as high-containment space even when no BSL-3 experiment is running. The proposed adaptable model aims to reserve those burdens for periods when the work requires them. The authors describe higher utility and operating costs in BSL-3 mode, while ordinary BSL-2 work can use a less intensive operating state.
That is a conditional advantage, not a proven financial result. The available design article does not quantify capital premiums, utility reductions, labor savings, downtime, conversion time or payback. A business case therefore needs project-specific energy modeling, staffing assumptions, maintenance pricing and an estimate of how often the suite will be placed in BSL-3 mode.
Begin with protocol-driven risk assessment
Before an architect draws a convertible room, the institution should identify the work it expects to perform. The risk assessment should describe agents, quantities, procedures, aerosols, animal or clinical materials, sharps, waste streams, exposure routes and the people who will enter the suite. It should also identify foreseeable future protocols rather than assuming that every possible BSL-3 activity will be accommodated.
The assessment becomes the design brief for the biosafety officer, principal investigators, facilities engineers, commissioning team and authorities having jurisdiction. It determines which BSL-3 features are needed, which are optional, and what must be demonstrated before use. The BMBL treats measures such as exhaust HEPA filtration and liquid effluent decontamination as dependent on the site-specific risk assessment; they should not be presented as automatic requirements for every facility labeled BSL-3.
Choose a location that can support containment
The proposed suite should sit in a controlled part of the building with limited entry and exit points. A secure location simplifies access control, visitor management, incident response and separation from unrelated traffic. Proximity to building mechanical infrastructure also matters: long, inaccessible service routes make inspection, filter work, valve maintenance and emergency intervention harder.

For new construction, an accessible mechanical floor above the laboratory can provide direct routes for supply and exhaust equipment. That arrangement consumes additional floor area and must be priced as part of the project. In a retrofit, an adjacent mechanical room or service zone may be more realistic, but the available space can constrain redundancy, duct routing and safe maintenance.
Use a room arrangement that supports both modes
The design article favors a controlled cluster of smaller rooms, potentially combining convertible laboratories with BSL-2 support rooms, rather than relying on one large open laboratory. A cluster can separate procedures, provide more than one work area and let the institution isolate a problem without shutting an entire floor. It also creates more doors, interlocks and pressure boundaries, so the flexibility is not free.
| Arrangement | Potential advantages | Design cautions |
|---|---|---|
| Clustered suite of smaller rooms | Separates procedures, supports staged access, offers multiple BSL-3-capable work areas and can keep BSL-2 support functions available. | Requires coordinated pressure relationships, more doors and controls, clear room-to-room decontamination procedures and careful maintenance access. |
| Large open laboratory | Can simplify furniture planning and routine BSL-2 circulation. | May provide fewer containment compartments and less operational flexibility when one procedure, equipment failure or decontamination event affects the room. |
Neither arrangement is universally superior. The right choice depends on the protocols, equipment, staffing pattern, decontamination method and the amount of work that must continue while one room is in BSL-3 mode.
Design personnel and material flows before drawing walls
Map every movement in both operating states: staff entry, changing, handwashing, equipment delivery, sample transfer, waste removal, maintenance access and emergency egress. The BSL-3 route should be deliberate and observable, with access limited to trained and authorized personnel.
A key planning question is whether BSL-2 users can reach their work areas independently while BSL-3 work is active. The proposed model considers separate or bypass access so routine traffic does not cross the containment suite’s controlled circulation. That route must not undermine pressure boundaries, security or emergency response. It also needs a written operating rule: who may use it, when it is locked, how deliveries are screened and how a simultaneous BSL-2 and BSL-3 schedule is approved.
Material flows deserve the same attention as people. Define clean and contaminated paths for incoming supplies, samples, reusable equipment and solid waste. Mark transfer points, staging areas and temporary storage locations on the plans. If a route changes between modes, the change should be visible in the room signage, access-control logic and standard operating procedure.

Make the mechanical system adaptable, not improvised
Provide access, control and redundancy
HVAC equipment, controls and service spaces should be reachable without sending maintenance staff through an active containment route whenever the risk assessment permits. Designers should evaluate redundant or adaptable air-handling capacity, controllable air valves, sensor locations, alarm logic and the consequences of a fan or control failure. The project team must define which components remain in service during a mode change and how the suite returns to a safe state after an interruption.
Filter housings, access doors and lifting paths need to be designed before equipment is installed. The article discusses placing filter housings before filters so the operating configuration can be changed when appropriate. It also proposes bypassing HEPA filtration in BSL-2 mode as an energy strategy. Those are author proposals, not universal specifications; the biosafety review must confirm whether a bypass is acceptable, how it is secured against incorrect operation and what testing is required after every change.
Do these 3 things before closing this tab:
1Clear out junk files and repair common Windows errors2Scan for outdated or missing drivers - takes under a minute3Repair Windows errors before they cause bigger problemsValidate ventilation in each operating state
Pressure relationships, directional airflow, supply and exhaust volumes, alarm set points and recovery behavior should be tested as an integrated system. The current BMBL guidance says ventilation performance is to be validated before the facility becomes operational, at least annually during operation and after significant ventilation modifications. The project should document exactly which tests demonstrate the approved design, who witnesses them and what happens when a result falls outside the acceptance range.
Validation is not a one-time handoff. Filter replacement, control-sequence changes, room reconfiguration, construction nearby or a major equipment failure can alter performance. The facility’s maintenance plan should trigger a biosafety review and, where applicable, repeat testing before the room returns to BSL-3 service.
Plan utilities, liquid effluent and solid waste
Water, power, emergency power, drainage, compressed gases, data and monitoring systems should be reviewed for both modes. Critical equipment may need dedicated circuits, backup power or alarm notification outside the suite. Utility shutoffs should be accessible to authorized responders without creating an uncontrolled entry.
Liquid effluent decontamination is a risk-assessment decision. The design article describes an effluent decontamination system (EDS) and a bypass concept as planning options. The BMBL likewise treats EDS installation as dependent on the site and program. If an EDS is selected, the institution must define treatment cycles, loading limits, monitoring, failure response and validation records. A bypass is not a shortcut around those decisions; it requires engineered isolation, documented authorization and a safe destination for the liquid.

Solid waste planning should place collection, staging and sterilization close enough to the suite to avoid unnecessary travel. The article highlights access to a sterilizer as an important consideration. The final procedure must specify container types, labeling, loading, cycle verification, cooling, spill response and what happens when a cycle fails. Waste requirements can differ by agent and jurisdiction, so the facility’s biosafety and environmental-health programs must approve them.

New construction
A new project can reserve a mechanical floor or service zone, align structural grids with containment rooms, place controlled entries at the perimeter and provide dedicated shafts. The trade-off is additional area and higher first cost. Early coordination can reduce later compromises because room boundaries, duct routes, electrical distribution and equipment replacement paths are established together.
Retrofit
A retrofit may use adjacent mechanical space and a compact suite, but existing floor-to-floor heights, structure, shafts, fire separations and occupied neighbors can limit options. Survey the building before committing to a conversion: verify available airflow, exhaust discharge locations, electrical capacity, drainage, emergency power, access routes and the ability to isolate construction dust and contaminants from occupied areas.
In either case, write the mode-change sequence before construction documents are complete. If the team cannot explain how the suite changes state, who authorizes it and how performance is verified, the design is not yet operationally complete.

Build a controlled changeover procedure
- Authorize the protocol. The principal investigator, biosafety officer and institutional review process confirm that the work and containment level are appropriate.
- Set the suite status. Access control, room signage, alarms, pressure targets and equipment settings are placed in the approved BSL-3 configuration.
- Verify the physical state. Trained staff check doors, interlocks, airflow indicators, cabinet certification status, decontamination supplies, emergency communications and waste equipment.
- Record the change. Log the operator, time, room, protocol, settings, test results and any deviation. Do not rely on an unrecorded verbal handoff.
- Operate and monitor. Keep required personnel and material routes active, respond to alarms under the incident plan and suspend work if a critical control is lost.
- Decontaminate and return. Follow the approved decontamination method, remove or treat waste, verify the room’s post-use condition and obtain authorization before restoring BSL-2 operation.
The exact sequence, test limits and sign-off roles are facility-specific. They should be exercised in drills before the first real BSL-3 experiment.
Questions to settle with the project team
- Which agents, quantities and procedures are in the design basis, and which future activities are explicitly excluded?
- Can BSL-2 users reach their work areas without entering an active BSL-3 circulation path?
- Where are the clean, contaminated, maintenance and emergency routes, and do they work in both modes?
- Which HVAC components are redundant, which are bypassable, and how is incorrect configuration prevented?
- Where can technicians replace filters, valves and controls without compromising containment?
- What ventilation tests are required before occupancy, annually and after modifications, and who accepts the results?
- Does the risk assessment require EDS, exhaust HEPA filtration or another treatment, and how will each system be validated?
- What happens when power, exhaust, pressure control, sterilization or monitoring fails?
- How often is BSL-3 operation expected, and what staffing and maintenance cost follows from that schedule?
- Which local codes, permits, occupational-health rules and institutional approvals apply in addition to BMBL guidance?
Common mistakes that undermine a conversion
- Designing only the room. Containment depends on the entire suite, including access, airflow, utilities, waste and procedures.
- Assuming a label changes performance. A BSL-3 sign does not establish pressure, filtration, decontamination or validated operation.
- Leaving maintenance until the end. Inaccessible fans, valves and filters turn routine service into a containment event.
- Treating HEPA or EDS as automatic. These choices must follow the protocol-specific risk assessment and be validated for the actual installation.
- Ignoring simultaneous users. Without an independent BSL-2 route and access rules, routine work can conflict with BSL-3 circulation.
- Promising savings without a model. Capital premiums, testing, staffing, energy, maintenance and downtime can outweigh an assumed operating benefit.
What can—and cannot—be claimed about cost efficiency
The adaptable-suite concept offers a plausible way to avoid running every experiment under the higher operating burden of BSL-3. It does not prove that every institution will save money. The published proposal contains no measured energy reduction, conversion schedule, lifecycle comparison or payback period, and no facility inspection or conversion test establishes those figures.
A defensible financial analysis should therefore compare at least two complete options: a continuously operated BSL-3 facility and a BSL-2 facility designed for controlled BSL-3 operation. Include construction and commissioning, mechanical redundancy, access control, certification, annual validation, filter and sterilizer maintenance, staffing, training, utilities, downtime during changeover and the expected number and duration of BSL-3 campaigns. Present the result as a range tied to stated assumptions, not as a guaranteed saving.
The Bottom Line
Designing BSL-2 space for verified BSL-3 operation can create useful capacity, but only when risk assessment, infrastructure, flows, validation and operating governance are designed as one system; the cost advantage must be demonstrated for the specific site.
